Cooper studeerde in 2000 af aan de London Academy of Music and Dramatic Art en speelde in de jaren daarna met name in het theater. Hij speelde in het stuk The History Boys (2004-2006) en werd hiervoor genomineerd voor een Drama Desk Award. Hij speelde ook in de verfilming van The History Boys (2006) en werd daarvoor genomineerd voor de British Independent Film Awards.
Sinds 2001 speelde Cooper een aantal kleinere filmrollen. Hij werd bij een breder publiek bekend door zijn rol als verloofde van Sophie (Amanda Seyfried) in de film Mamma Mia!. Samen met Sophie zong hij in de film het ABBA-nummer Lay All Your Love on Me.

She gained international recognition for her portrayal of Mildred Loving in the Jeff Nichols directed historical romance drama Loving (2016), earning a nomination for the Academy Award for Best Actress and the BAFTA Rising Star Award.
She played an African-American woman passing as a white woman in the Rebecca Hall directed period drama Passing (2021), earning nominations for the BAFTA Award for Best Actress in a Supporting Role and Golden Globe Award for Best Supporting Actress – Motion Picture. Negga has also appeared in the films Isolation (2005), Breakfast on Pluto (2005), Warcraft (2016), Ad Astra (2019) and Good Grief (2023).
